Nuclear Factor kappa-light-chain-enhancer of activated B cells, commonly abbreviated as NF-κB, is a protein complex that functions as a transcription factor and is pivotal in regulating the expression of various genes that are crucial for a range of cellular processes. NF-κB inhibitors comprise a chemical class designed to impede the activity of the NF-κB pathway. Typically, these inhibitors target various points within the signaling cascade that activates NF-κB, which is known for its role in controlling the transcription of DNA, cytokine production, and cell survival. The NF-κB complex is kept in an inactive state in the cytoplasm under normal conditions, bound to inhibitory proteins known as IκBs. In response to specific external stimuli, the IκB proteins become phosphorylated, leading to their ubiquitination and subsequent proteasomal degradation, which then allows NF-κB to translocate into the nucleus and initiate gene transcription.
