Chemical activators of Olr559 can initiate various intracellular pathways leading to its functional activation. Forskolin directly stimulates adenylyl cyclase, elevating intracellular cAMP levels, which then activates protein kinase A (PKA). PKA can phosphorylate target proteins, and in this cascade, Olr559 can be functionally activated. Similarly, PMA, known as phorbol 12-myristate 13-acetate, activates protein kinase C (PKC), another kinase capable of phosphorylating Olr559, thereby facilitating its activation. Ionomycin, by increasing intracellular calcium levels, can activate calcium-dependent kinases that may phosphorylate and thus activate Olr559. This elevation in calcium levels can also be achieved through BAY K8644, an L-type calcium channel agonist, which could lead to the activation of Olr559 by downstream calcium-stimulated kinases.
Moreover, Zinc Pyrithione raises intracellular zinc levels, which can activate zinc-sensitive kinases, leading to the phosphorylation and activation of Olr559. Okadaic Acid, by inhibiting protein phosphatases, prevents the dephosphorylation of proteins, thus maintaining Olr559 in an active phosphorylated state. Contrary to the typical function of PKA inhibitors, H-89 Dihydrochloride can, under some circumstances, lead to the activation of Olr559 by non-PKA kinases compensating for inhibited PKA. Calyculin A, like Okadaic Acid, inhibits protein phosphatases such as PP1 and PP2A, resulting in a sustained phosphorylated and active state of Olr559. Anisomycin activates stress-activated protein kinases, which in turn can activate Olr559 by phosphorylation. Veratridine, by acting as a sodium channel agonist, could initiate a cascade involving sodium-sensitive kinases that phosphorylate and activate Olr559. Thapsigargin, a SERCA inhibitor, leads to increased cytosolic calcium, which activates calcium-dependent kinases that can then phosphorylate and activate Olr559. Finally, Ouabain, by inhibiting the Na+/K+ ATPase, alters intracellular ion gradients, which could lead to the activation of Olr559 through changes in kinase activity associated with altered ion balance.
