NOR-1 inhibitors, direct or indirect, are chemical compounds that are capable of reducing the activity of the NOR-1 protein. Several compounds can indirectly impact NOR-1 activity by modulating the activity of various upstream signaling pathways. By inhibiting PI3K or mTOR, these compounds can reduce the activation of AKT, a protein kinase that can phosphorylate and activate NOR-1. Thus, the inhibition of PI3K or mTOR can indirectly lead to reduced NOR-1 activity. These compounds inhibit kinases like MEK or c-RAF, which are involved in the activation of ERK. Other compounds, such as AG-490 and PP2, target the JAK/STAT pathway. These inhibitors can prevent the activation of STAT3, a transcription factor that can increase NOR-1 expression. Finally, BAY 11-7082 inhibits NF-kB, a transcription factor that can activate AP-1, which can increase NOR-1 expression. By inhibiting NF-kB, this compound can indirectly reduce NOR-1 levels.
