Chemical activators of Mucin 7 (MUC7) can induce a range of intracellular signaling cascades leading to the functional activation of this protein. Calcium chloride, for instance, raises intracellular calcium levels, which can activate calcium-dependent protein kinases. These kinases, in turn, are capable of phosphorylating MUC7, thereby prompting its activation. Similarly, phorbol 12-myristate 13-acetate (PMA) engages protein kinase C (PKC). Activated PKC can then directly phosphorylate MUC7 on specific serine or threonine residues, resulting in the protein's functional activation. Another activator, Forskolin, functions by escalating cAMP levels within the cell, which activates protein kinase A (PKA). PKA can phosphorylate MUC7, leading to its functional activation. The synthetic DAG analog, 1,2-Dioctanoyl-sn-glycerol (DiC8), also activates PKC, which can phosphorylate and activate MUC7. Ionomycin, by increasing intracellular calcium concentration, can activate calcium-dependent kinases that may target MUC7 for phosphorylation and subsequent activation.
Continuing with other chemical activators, Dibutyryl cAMP, a cell-permeable analog of cAMP, activates PKA, and this can result in the phosphorylation and activation of MUC7. The calcium ionophore A23187 elevates intracellular calcium levels, which can activate kinases that phosphorylate and activate MUC7. Thapsigargin disrupts calcium stores in the endoplasmic reticulum, resulting in a rise in cytosolic calcium that may activate kinases that phosphorylate MUC7. Bryostatin 1 modulates PKC activity, leading to phosphorylation and activation of MUC7. Okadaic acid, an inhibitor of protein phosphatases, can lead to increased phosphorylation levels of several proteins, possibly including MUC7, which would result in its activation. Hydrogen peroxide, through oxidative stress, can modulate signaling pathways that lead to the activation of kinases that phosphorylate MUC7. Lastly, the nitric oxide donor SNAP, by raising cGMP levels, can activate protein kinases that may phosphorylate and activate MUC7, showcasing the diverse chemical interactions that can lead to the functional activation of this mucin protein.
