Chemical activators of MAST205 include a spectrum of compounds that directly or indirectly promote the phosphorylation state of the protein, a common mechanism for functional activation. Forskolin and 1,9-Dideoxyforskolin both act on adenylate cyclase to increase intracellular cAMP, which in turn activates PKA. PKA has a wide substrate range and can phosphorylate MAST205, leading to its activation. Similarly, PMA operates through the activation of PKC which has the ability to directly phosphorylate MAST205. Ionomycin and A-23187 Calcium Ionophore elevate intracellular calcium levels, which can activate calcium-dependent kinases such as CaMK; these kinases can phosphorylate MAST205, resulting in its activation. Thapsigargin works by increasing cytosolic calcium through inhibition of the SERCA pump, again leading to activation of calcium-sensitive kinases that may target MAST205.
Compounds like Okadaic Acid and Calyculin A inhibit protein phosphatases, thus preventing dephosphorylation and maintaining proteins like MAST205 in their phosphorylated, active form. Staurosporine, although a kinase inhibitor, can at low doses activate certain kinases that may phosphorylate MAST205. KN-93, as an inhibitor of CaMKII, may lead to a compensatory upregulation of alternative kinases that can phosphorylate and activate MAST205. Bisindolylmaleimide I, known primarily as a PKC inhibitor, may also induce a compensatory activation of other kinases which can lead to MAST205 activation. Anisomycin, through its activation of MAPK pathways, can lead to the activation of MAST205. Collectively, these chemicals act through various pathways to influence the phosphorylation status of MAST205, culminating in its functional activation.
