KLHL12 Activators are chemicals that can increase the functional activity of KLHL12, a protein that plays a vital role in the ubiquitination of G proteins and is involved in cAMP signaling and Protein Kinase C (PKC) pathway. Compounds like Forskolin, a potent adenylate cyclase activator, and Rolipram, a selective phosphodiesterase-4 inhibitor, enhance cAMP production, indirectly increasing the interaction of KLHL12 with its G protein substrates. Similarly, IBMX, another phosphodiesterase inhibitor, and ZM241385, an antagonist of the adenosine A2A receptor, work by enhancing cAMP levels, which in turn facilitates KLHL12's functional activity. Furthermore, SQ22536, an adenylate cyclase inhibitor, and KT5720, a selective inhibitor of Protein Kinase A (PKA), modulate cAMP levels, thereby indirectly augmenting the functional activity of KLHL12.
In addition, several KLHL12 activatorsinteract with the Protein Kinase C (PKC) pathway, which is known to enhance the phosphorylation and activation of KLHL12. PMA, a potent activator of PKC, and Go6976 and BIM I, both selective PKC inhibitors, can indirectly increase the demand for KLHL12's role in the PKC pathway, thereby enhancing its functional activity. Moreover, Oxotremorine-M, Atropine and Pilocarpine are muscarinic acetylcholine receptor (mAChR) agonists or antagonists that can enhance the phosphorylation and activation of KLHL12. The activation of mAChR can lead to an increased demand for KLHL12's role in this pathway.
