IL-17RE Activators

Interleukin-17 receptor E (IL-17RE) is an integral component of the IL-17 receptor family, holding significance in the orchestration of immune responses. The family of chemicals known as IL-17RE Activators comprises various compounds that can indirectly influence the function and signaling of IL-17RE. These compounds typically target distinct cellular pathways closely associated with IL-17RE activity, thus serving as indirect modulators of its functionality.

Among the notable compounds, Phorbol 12-myristate 13-acetate (PMA) stands out as a potent activator of protein kinase C (PKC), which is intricately linked to IL-17 signaling pathways. U0126, a selective inhibitor for MEK1 and MEK2, along with SB203580, a p38 MAPK inhibitor, can modify the signaling cascades emanating from IL-17RE. The phosphoinositide 3-kinases (PI3K) inhibitors, Ly294002 and Wortmannin, affect the Akt pathway, thereby influencing IL-17 responses. Furthermore, SP600125, a JNK inhibitor, and BAY 11-7082, which impedes NF-κB activation, directly influence downstream pathways of IL-17 signaling. Cyclosporin A can exert an effect on IL-17 producing cells, thereby indirectly influencing IL-17RE, while Ionomycin, as a calcium ionophore, plays a role in elevating intracellular calcium levels and modulating numerous signaling pathways. Lastly, IKK Inhibitor VII and Anisomycin act on the IKK and JNK pathways respectively, both of which are implicated in IL-17 mediated signaling.
