Wortmannin is an antifungal antibiotic(similar to viridin) and a fungal metabolite. It is a potent and selective inhibitor of phosphatidylinositol 3-kinase. Wortmannin inhibits repair of DNA double-strand breaks in irradiated normal human cells. Wortmannin was also shown to have enhancing influence on memory and impair learning abilities. It has an in vitro inhibitory concentration (IC50) of around 5 nM, making it a more potent inhibitor than LY294002, another used PI3K inhibitor. Wortmannin displays a similar potency in vitro for the class I, II, and III PI3K members although it can also inhibit other PI3K-related enzymes such as mTOR, DNA-PK, some phosphatidylinositol 4-kinases, myosin light chain kinase (MLCK) and mitogen-activated protein kinase (MAPK) at high concentrations. Wortmannin has also been reported to inhibit members of the polo-like kinase family with IC50 in the same range as for PI3K. The half-life of Wortmannin in tissue culture is about 10 minutes due to the presence of the highly reactive C20 carbon that is also responsible for its ability to covalently inactivate PI3K.

Wortmannin is said to be unstable in cell culture media ( t1/2 of 10 mins). How can I make serial dilutions in media and use this to treat cells in culture without losing its activity?

Thank you for your question. Unfortunately, we are not able to find any recommended cell culture protocol for use of Wortmannin, and we have not used it in our own cell culture labs. We did find a paper that may serve as a useful reference, please see PMID: 14622954.
