GSDML Activators

GSDML Activators encompass a broad range of chemicals that can indirectly influence the function of the GSDML protein or the intricate pathways with which it interacts. Originating from diverse chemical backgrounds, these activators function by affecting different signaling cascades and cellular processes that are connected, either directly or indirectly, to GSDML's role in the cell.

A quintessential example is Phorbol 12-myristate 13-acetate (PMA), a potent activator of Protein Kinase C (PKC). By enhancing PKC activity, PMA can influence various downstream signaling events associated with pyroptosis, the form of cell death in which GSDML plays a pivotal role. Similarly, Nigericin, a potassium ionophore, is recognized for its ability to activate NLRP3 inflammasomes, presenting another pathway that can intersect with GSDML's function. A subset of these activators, including U0126, SB203580, Ly294002, and SP600125, target the intricate web of MAPK signaling pathways. These pathways, comprising ERK, p38, and JNK cascades, hold importance in various cellular responses, including those linked to pyroptosis. On the other hand, BAY 11-7082's mechanism revolves around the inhibition of NF-κB activation, showcasing another potential route of influence on GSDML-associated pathways. Furthermore, agents like Cyclosporin A modulate T-cell activation, offering yet another layer of indirect interaction. Lastly, chemicals like Ionomycin, which perturb intracellular calcium levels, or MG-132, which interfere with protein turnover by inhibiting proteasomes, underscore the varied mechanisms through which these activators can intersect with the functional realm of GSDML.
