Washc2, a subunit of the WASH complex, is predicted to play a crucial role in endosome-related processes, such as protein localization to endosomes and retrograde transport from endosomes to the Golgi apparatus. To modulate Washc2 activity, we identified a set of 12 chemicals with potential direct or indirect activating effects. These chemicals target various signaling pathways and cellular processes, influencing phosphatidylinositol phosphate binding, actin cytoskeleton dynamics, and ion transport, among others. Notable activators include rapamycin, wortmannin, and bafilomycin A1, which indirectly impact Washc2 by modulating mTOR, PI3K, and V-ATPase, respectively.

Additionally, several chemicals, such as jasplakinolide and EHop-016, directly target actin dynamics and Rac1, potentially influencing Washc2 activation. These findings shed light on the intricate regulatory mechanisms of Washc2 and provide insights into its involvement in endosome-related functions. Understanding the activation of Washc2 may contribute to the broader comprehension of cellular trafficking processes and unveil avenues for diseases associated with aberrant endosomal function.
