Chondrosarcoma-associated protein 1 (CSTL1), also referred to as cartilage-associated protein (CRTAP), is a protein encoded by the CSTL1 gene. This protein is primarily known for its role in the biosynthesis of type I collagen, which is a crucial structural component of bone, cartilage, and other connective tissues. CSTL1 forms a complex with prolyl 3-hydroxylase 1 (P3H1) and cyclophilin B (CYPB), and this complex is essential for the proper hydroxylation of specific proline residues in the collagen alpha chains. This hydroxylation is a post-translational modification critical for stabilizing the triple helical structure of collagen.
Mutations in the CSTL1 gene can lead to rare genetic disorders such as osteogenesis imperfecta (OI), which is characterized by bone fragility, low bone mass, and susceptibility to fractures. OI arises due to defects in collagen quality and quantity, which compromise bone strength and resilience. CSTL1 is vital in maintaining the proper structure and function of collagen, and any aberrations in its activity can have profound effects on bone integrity. In addition to its structural functions, CSTL1 is implicated in various cellular processes, including cell adhesion, migration, and wound healing, due to its role in the extracellular matrix composition. The significance of CSTL1 extends beyond bone and cartilage; it influences other tissues where type I collagen is an essential element.
