caspase-2 Antibody (F-2) is a mouse monoclonal IgG1 kappa light chain antibody that detects caspase-2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-caspase-2 antibody (F-2) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Caspase-2, also known as Nedd2 or ICH-1, is an aspartate-specific cysteine protease that plays a critical role in the apoptotic process, being activated in response to various apoptotic stimuli. Notably, caspase-2 is unique among caspases due to its dual characteristics, possessing features of both upstream caspases with a long prodomain and downstream caspases with DEXD substrate specificity. Caspase-2 is predominantly localized in the mitochondria, Golgi apparatus, cytoplasm, and nucleus, which is essential for its function in regulating apoptosis. Caspase-2 exists in two isoforms, caspase-2L and caspase-2S, produced through alternative splicing, with caspase-2L acting as a positive regulator of apoptosis and caspase-2S serving as a negative regulator. Upon apoptotic stimuli, caspase-2L precursor is cleaved at Asp 153, resulting in the formation of the p30 fragment, which can undergo further cleavage to yield the p18 and p13/p14 fragments. As apoptosis progresses, p13/p14 fragment can be processed into the p12 fragment, highlighting the intricate regulatory mechanisms of caspase-2 in cell death pathways.
