Thank you for your question. This secondary antibody reagent, m-IgGκ BP-HRP: sc-516102 can be used with any mouse monoclonal primary antibody. I would suggest testing it on maybe a monoclonal loading control primary antibody such as actin or GAPDH to make sure you see the HRP signal.

In Datasheet appears a microphotograph of ERa (C-3) antibody detection and legend indicates that primary antibody is SC-514857, however, the antibody is not currently enlisted in Santa Cruz catalog. Could you send me information about SC-514857 antibody?

Thank you for your question. Yes, you can use this mouse IgG binding protein secondary reagent for ELISA as long as your primary antibody is appropriate for this reagent (i.e. kappa light chain).

Can this secondary antibody be used for mouse tissue homogenate to do Western Blot? If not, are there any suggestions? I have quite a few primary antibodies from Santa Cruz and they are all mouse antibodies. I am hoping to be still able to use them.

Thank you for your question. Yes, this secondary antibody can be used for Western blot detection of mouse monoclonal primary antibodies (with a kappa light chain) on mouse tissue extracts. We use it routinely in our labs and it works fantastic! It is more specific than most conventional polyclonal anti-mouse IgG secondary antibodies (e.g., donkey anti-mouse IgG, etc.) since it specifically binds to only the kappa light chain of mouse IgG. We always advise including a secondary control (omit primary antibody) with all of your blots just to identify any possible non-specific binding of your secondary antibody alone.

I would like to detect GST-tagged proteins by WB. Is the Sc-516102 the correct ab and I do not require a secondary ab for ECL-HRP? Thanks Michael

Thank you for your question. We offer a variety of GST-specific primary antibodies. Our GST Antibody (B-14): sc-138 in particular has been featured in several hundreds of publications and is a reliable research tool. For secondary detection with HRP we recommend our secondary binding protein m-IgGκ BP-HRP: sc-516102. Alternatively, the sc-138 HRP direct conjugate can be used. This product does not require secondary detection step, as it is already conjugated with HRP.

If I were to combine something like sc-271164 as my primary andtibody and the m-IgGκ BP-HRP as my secondary (based on the specifics of the antibodies binding) for western blot, what chemiluminescence would you recommend? Would it work with west femto?

Thank you for your question. I would not recommend using two secondaries, i.e. using both HRP conjugated secondary and a chemiluminescence secondary. I would just select a secondary antibody with a fluorochrome. You can find them here: https://www.scbt.com/scbt/browse/Secondary-Antibodies/_/N-1hoaxk4 Our HRP conjugated secondary should be fine with West Femto, but not in combination with a fluorchrome conjugated antibody. Does this binding protein bind to denatured antibody (ie. boiled and reduced IP eluate), or does it only bind to native antibody.

Thank you for your question. This will recognize the light chain of mouse IgG under reducing conditions (e.g., if you have boiled and reduced a mouse monoclonal IgG primary antibody used in IP).

Thank you for your question. This is not raised in a host animal like a conventional secondary antibody. This is a recombinant protein that specifically binds to only the kappa light chains of mouse IgG and mouse IgM.
