Hi, regarding m-IgGkBP-HRP (SC-516102), May I know the recommended starting dilution for western blot? Can I see the result by the naked eye? or What is reagents that I have to use to observe the bands? Can you introcduct to me some reagents?

Thank you for your question. As indicated in the product datasheet, the recommended starting dilution is 1:1000, with a dilution range of 1:1000-1:10000. Our recommended support products for visualization is Western Blotting Luminol Reagent sc-2048. Luminol requires a device for detection of chemiluminescence.

Would this protein bind to human IgG kappa light chain present in the sample and cause non-specific signals?

I'm using this product for detection of COX-2 and HIF-1a primary AB (Santa Cruz as well), in recommended dilution. Still, have too many unspecific bindings. My blocking buffer is 5% nonfat milk with TBST, blockage time 1h.

Can I use Mouse IgG kappa Binding Protein conjugated to HRP for double-immunostainig? Does this Binding Protein recognize other animal IgG such as rabbit or Goat?

Thank you for your question. Mouse IgG kappa Binding Proteins recognize any mouse IgG that possess a kappa light chain (IgG, IgA, IgM). It should not cross-react with Ig of other species such as rabbit or goat.

Can this product be used for detection of mouse blood IgG by ELISA?

Thank you for your question. m-IgGκBP-HRP is recommended for immunohistochemistry (including paraffin-embedded sections) at a starting dilution: 1:25, dilution range 1:25-1:100. Optimal dilution to be determined by titration. For complementary reagents to complete your staining protocol, see also: UltraCruz® Blocking Reagent (sc-516214), Hydrogen Peroxide (sc-203336) and DAB, 50X (sc-24982).
