caspase-2L Antibody (F-7) is a mouse monoclonal IgG1 kappa light chain antibody that detects caspase-2L protein, including the p13 subunit and full-length caspase-2 precursor, in mouse, rat, and human samples through western blotting, immunoprecipitation, immunofluorescence, and enzyme-linked immunosorbent assay. caspase-2L Antibody (F-7) is offered in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase, phycoerythrin, fluorescein isothiocyanate, and multiple Alexa Fluor® conjugates. Caspase-2, also known as Nedd2 or Ich-1, is an aspartate-specific cysteine protease that plays a crucial role in the apoptotic process, being activated in response to various apoptotic stimuli. Caspase-2 is unique among caspases due to its dual characteristics, possessing features of both upstream caspases, with a long prodomain, and downstream caspases, with DEXD substrate specificity. Notably, caspase-2 is predominantly expressed in the brain during development and is found at lower levels in adult tissues. Caspase-2 is localized in several cellular compartments, including the mitochondria, Golgi apparatus, cytoplasm, and nucleus, which is significant as localization is essential for its role in apoptosis regulation. Caspase-2 exists in two isoforms, caspase-2L and caspase-2S, generated through alternative splicing, with caspase-2L acting as a positive regulator of apoptosis and caspase-2S serving as a negative regulator. Upon apoptotic signals, the caspase-2L precursor is cleaved at Asp 153, resulting in the formation of the p30 fragment, which is further processed to yield the p18 fragment and the p13 or p14 fragment. As apoptosis progresses, the p13 (or p14) fragment can undergo additional processing to produce the p12 fragment, highlighting the complex regulatory mechanisms of apoptosis in which caspase-2 plays a pivotal role.
