Suppressor of Hairless inhibitors are chemicals that have the ability to indirectly reduce the function or expression of the Suppressor of Hairless (Su(H)) protein, a crucial transcription factor in the Notch signaling pathway. These inhibitors operate by targeting different signaling pathways that interact with the Notch pathway, where Su(H) plays a central role. Inhibitors such as U0126 and PD98059 target the MEK1/2 proteins in the MAPK/ERK pathway, reducing ERK phosphorylation and indirectly reducing Su(H) activity. Similarly, LY294002 and Wortmannin inhibit the PI3K protein in the PI3K/Akt pathway, thereby suppressing Notch signaling and Su(H) activity. SP600125 and AG490 target the JNK and JAK2 proteins, respectively, in the JNK and JAK/STAT pathways. By inhibiting these proteins, these chemicals can reduce Notch signaling, leading to decreased activity of Su(H). Flavopiridol, a CDK inhibitor, can destabilize Su(H) by preventing its phosphorylation and stabilization.
Rapamycin, an mTOR inhibitor, and SB431542, a TGF-beta pathway inhibitor, can suppress Notch signaling, and hence Su(H) activity, by inhibiting the mTOR and TGF-beta pathways, respectively. Cyclopamine, a Hedgehog pathway inhibitor, can also suppress Su(H) activity by inhibiting the Hedgehog pathway, which interacts with the Notch pathway. DAPT and RO4929097, both gamma-secretase inhibitors, function by inhibiting the cleavage of Notch and the subsequent release of NICD, which interacts with Su(H). By inhibiting gamma-secretase, these chemicals can suppress Su(H) activity. Therefore, while these inhibitors do not directly target Su(H), they can indirectly modulate its activity by influencing the function of other proteins and signaling pathways that interact with the Notch pathway. The convergence and crosstalk of these pathways at the level of Su(H) demonstrates its central role in various cellular processes.
