NTN Activators are a group of chemicals that can enhance the functional activity of NTN by primarily inhibiting or activating the activity of various signaling molecules that are known to directly or indirectly affect NTN. These molecules include NF-κB, multiple kinases, SIRT1, TGF-β receptor, adenylate cyclase (which affects cAMP and PKA), AKT, antioxidants to reduce oxidative stress, soluble guanylyl cyclase (which affects cGMP and PKG), A2A adenosine receptor, and calcium/calmodulin-dependent protein kinase II (CaMKII). By influencing these molecules, the NTN Activators can significantly enhance the functional activity of NTN.
Curcumin and Resveratrol enhance NTN activity by inhibiting NF-κB and activating SIRT1, respectively, thereby reducing NF-κB-mediated inflammatory responses and promoting the deacetylation and activation of NTN. Quercetin and Genistein act as broad-spectrum kinase inhibitors, thereby potentially increasing NTN signaling by inhibiting kinases that downregulate NTN. SB431542 enhances NTN activity by selectively inhibiting the TGF-β receptor and reducing TGF-β-mediated inhibition of NTN. Forskolin, YC-1, and Puerarin enhance NTN activity by activating adenylate cyclase and soluble guanylyl cyclase and AKT, respectively, leading to increased NTN activation through the phosphorylation of NTN. EGCG enhances NTN activity by acting as a potent antioxidant and reducing oxidative stress that attenuates NTN expression and signaling. H-89, ZM-241385, and KN-62 enhance NTN activity by inhibiting PKA, A2A adenosine receptor, and CaMKII, respectively, thereby preventing PKA-dependent phosphorylation of NTN, blocking adenosine-dependent inhibition of NTN, and preventing CaMKII-dependent phosphorylation of NTN.
