Matriptase Inhibitors

Matriptase Inhibitors, as a class, include a variety of chemical compounds that effectively inhibit the serine protease activity of matriptase. Matriptase plays a significant role in epithelial barrier maintenance, innate immunity, and pathological processes, including cancer progression. The inhibitors such as Nafamostat mesylate, SBTI, AEBSF, and Camostat mesylate, are primarily serine protease inhibitors. Nafamostat mesylate is a synthetic inhibitor with a broad spectrum of action, known for its effective inhibition of matriptase. SBTI, found in soybeans, while traditionally known as a trypsin inhibitor, has shown activity against matriptase. AEBSF and Camostat mesylate are also effective against a range of serine proteases, including matriptase.

Other inhibitors like Gabexate mesylate and Benzamidine are broad-spectrum serine protease inhibitors, known for their efficacy in inhibiting matriptase among other proteases. Aprotinin, a bovine lung-derived inhibitor, and Leupeptin, a more general serine protease inhibitor, also exhibit inhibitory activity against matriptase. In addition to the primary serine protease inhibitors, compounds like E-64, Pepstatin A, Phosphoramidon, and Marimastat, though targeting other protease classes (cysteine, aspartic, and metalloproteases, respectively), have been found to exhibit some degree of inhibition towards matriptase. This cross-reactivity highlights the intricate aspects of protease inhibition and suggests that broad-spectrum inhibitors may be capable of targeting various protease types, such as matriptase.
