HRASLS5 can initiate various intracellular signaling pathways leading to its phosphorylation and subsequent activation. Phorbol 12-myristate 13-acetate and PMA are both phorbol esters that directly activate protein kinase C (PKC). Once activated, PKC can phosphorylate HRASLS5. Similarly, Forskolin and Isoproterenol function through the elevation of cyclic AMP (cAMP) levels, with Forskolin directly activating adenylyl cyclase and Isoproterenol acting as a beta-adrenergic agonist. The rise in cAMP levels activates protein kinase A (PKA), which then can phosphorylate and activate HRASLS5. Dibutyryl-cAMP, a cAMP analog, bypasses the receptor-mediated activation and directly activates PKA, leading to a similar phosphorylation event on HRASLS5. Additionally, ionomycin and A23187, both calcium ionophores, can increase intracellular calcium concentrations, which can activate calcium-dependent protein kinases capable of modifying HRASLS5.
Epidermal Growth Factor (EGF) stimulates its receptor EGFR, triggering a cascade that involves MAPK/ERK kinases, which can phosphorylate HRASLS5. Anisomycin, which is known to activate MAPK/ERK kinases, can similarly lead to the phosphorylation of HRASLS5. Okadaic acid, by inhibiting protein phosphatases 1 and 2A, prevents the dephosphorylation of proteins, which may result in the sustained phosphorylation and activation of HRASLS5. Bryostatin 1, as a modulator of PKC, can also result in the phosphorylation of HRASLS5 through PKC's kinase activity. Thapsigargin contributes to HRASLS5 activation by inhibiting the sarco/endoplasmic reticulum Ca2+-ATPase (SERCA), causing an increase in cytosolic calcium levels which then can activate kinases that phosphorylate HRASLS5. All these chemicals, through distinct mechanisms, converge on the activation of HRASLS5 via phosphorylation.
