Fibroblast Growth Factor 12 (FGF-12), also known as FGF homologous factor 2 (FHF2), is a protein encoded by the FGF12 gene in humans. While it's part of the fibroblast growth factor (FGF) family, FGF-12 doesn't function like typical FGFs, which are generally involved in cell growth, tissue repair, and angiogenesis. Instead, FGF-12 is classified as an 'intracellular' or 'non-secreted' FGF.FGF-12 lacks the conventional FGF receptor-binding domain and doesn't activate FGF receptors. It is primarily localized to the intracellular environment, where it interacts directly with the voltage-gated sodium channels (NaV). These channels are crucial for generating and propagating action potentials in neurons and muscle cells.
FGF-12 is known to modulate the function of these channels, affecting the electrical excitability of the cells. It binds to the α-subunit of the sodium channel, modulating its gating kinetics and voltage dependencies. This makes FGF-12 important in nervous system function and potentially in conditions relating to electrical signal transmission, such as certain neurological disorders and cardiac conditions.Studies have suggested a role for FGF-12 in the development and function of the nervous system, particularly the brain. Alterations in FGF-12 expression or function could potentially contribute to neurological diseases, although the exact mechanisms are still being explored.
