ARHGAP15 Inhibitors

ARHGAP15 inhibitors primarily function by impeding Rho GTPase signaling. This diverse range of chemicals can potentially influence the activation, post-translational modification, and downstream effects of Rho family GTPases. Y-27632 and Fasudil, for example, serve as Rho kinase (ROCK) inhibitors, which can suppress RhoA signaling and its associated stress fiber formation. Similarly, Rhosin acts as a direct barrier to RhoA GTPase.

Compounds such as NSC 23766 and EHT 1864 focus on the Rac arm of the Rho GTPases. While NSC 23766 thwarts Rac1-GTP from binding to its effector PAK1, EHT 1864 ensures Rac remains in its dormant GDP-bound state. Further targeting the GEF-GTPase interaction, ITX3 inhibits Trio, a RhoGEF, which obstructs RhoG activation. ML141 and ZCL278 concentrate on the Cdc42 GTPase. Both chemicals act as inhibitors, with ML141 averting Cdc42 activation and ZCL278 hampering the interaction between Cdc42 and its specific GEFs. Additionally, GGTI-298, a Geranylgeranyltransferase I inhibitor, perturbs the post-translational modification of Rho proteins, indirectly affecting their activity.Lastly, Latrunculin A, an actin polymerization inhibitor, provides an indirect approach by hindering downstream signaling associated with Rho GTPases, emphasizing the intertwined relationship between actin dynamics and Rho GTPase signaling.
