Similarly, compounds like Forskolin and 8-Bromo-cAMP, which elevate intracellular cAMP levels, activate protein kinase A (PKA). PKA, in turn, phosphorylates ANKMY1. Dibutyryl-cAMP (db-cAMP) also plays a similar role by entering the cell and directly activating PKA, which can then target ANKMY1 for phosphorylation. Ionomycin and A-23187, both calcium ionophores, increase intracellular calcium concentrations. This rise in calcium can activate calmodulin-dependent protein kinases that have the capacity to phosphorylate ANKMY1. Additionally, 4α-Phorbol 12,13-didecanoate, another phorbol ester, activates certain PKC isoforms, which can then phosphorylate ANKMY1.
