ANGEL2 Inhibitors

Compounds like AKT inhibitor VIII, Perifosine, Triciribine, GSK690693, MK-2206, and API-2 directly inhibit the Akt signaling pathway. Akt, also known as protein kinase B, is a serine/threonine-specific protein kinase that plays a key role in multiple cellular processes such as glucose metabolism, apoptosis, cell proliferation, transcription, and cell migration. By directly inhibiting Akt, these compounds can decrease the phosphorylation and activation of ANGEL2. Furthermore, Rapamycin and PI-103 can disrupt the mTOR signaling downstream of Akt. mTOR is a serine/threonine protein kinase that regulates cell growth, cell proliferation, cell motility, cell survival, protein synthesis, and transcription. By inhibiting mTOR, these compounds can further reduce the activation of ANGEL2. The result of these combined actions is a significant reduction in the functional activity of ANGEL2, demonstrating the effectiveness of these specific inhibitors on the PI3K/Akt signaling pathway and its associated proteins.

The specificity of these inhibitors for the PI3K/Akt pathway and the subsequent effect on ANGEL2 is not entirely coincidental. Given the pivotal role that this pathway plays in a myriad of cellular processes, it is logical that the inhibition of key components would have downstream effects on related proteins, such as ANGEL2. The specific biochemical actions of these inhibitors on PI3K, Akt, and mTOR, and the subsequent reduction in ANGEL2 activity, reflect a clear and direct modulation of the functional state of ANGEL2.
