Topo I Antibody (C-21) is a mouse monoclonal IgM antibody that detects Topo I protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-Topo I antibody (C-21) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Topo I protein, also known as DNA topoisomerase I, plays a crucial role in maintaining DNA structure integrity during cellular processes such as replication, transcription, and recombination. By introducing a transient break in one strand of the DNA helix, Topo I alleviates torsional strain that arises ahead of the replication fork, preventing DNA supercoiling and ensuring proper DNA function. This enzymatic activity is vital for cellular health, as dysregulation of Topo I can lead to genomic instability and has been implicated in various cancers, making Topo I a significant target for anticancer therapies. Topo I interacts with several other proteins, including transcription factors and repair proteins, which further underscores Topo I′s importance in cellular processes and highlights Topo I′s potential as a therapeutic target.

What is the position of the epitope of Topoisomerase I against which this antibody is raised??

Thank you for your question. This Topoisomerase antibody was raised against recombinant Topo I of human origin. The exact epitope has not been mapped.

Why there are more than two forms? Are they two isoforms of TOPO1 beta ?

Thank you for your question. The expected molecular weight for Topo 1 is ~91-100kDa. This is dependent on glycosylation as it is a highly glycosylated protein. Does Topo I (c-21) sc-32736 work well in RIP/CLIP experiments, using standard protein A/G beads?

Thank you for your question. We have not tested this mouse monoclonal IgM, Topo I Antibody (C-21): sc-32736, for RIP or CLIP experiments, and so those applications are not recommended at this time. This antibody is recommended for WB, IP, IF and IHC(P).

Can Topo I (C-21): sc-32736 be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Thank you for your question. Yes, Topo I (C-21): sc-32736 has been validated for use in IHC with paraffin-embedded sections. We performed staining for TOP1 both in PFA fixed and Triton X-100-permeabilized cells or MeOH-fixed cells usin the range of the dilutions - from 1:50 to 1:500 - no success at all

Rated 3 out of 5 by from Staining is strong, location is not precisewe used this antibody for our IHC stain on tumour sample, the TOPOⅠ protein located in cell nucleus. In our tissue slice, the nucleus is positive, colour is strong, but at the same time, cytoplasm is positive too.
