Smad7 Antibody (B-8) is a mouse monoclonal IgG1 kappa light chain antibody that detects Smad7 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-Smad7 antibody (B-8) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Smad7 protein plays a crucial role in regulating TGFβ (transforming growth factor beta) signaling pathways, acting as a negative regulator that inhibits phosphorylation and activation of other Smad proteins, particularly Smad2 and Smad3. I've used the Smad7 antibody (sc-365846) and detected two bands about 30kDa and 75kDa. I've examined mouse kidney tissue that was homogenized using RIPA lysis buffer. Do you have information about the two detected bands?

Thank you for your inquiry and your interest in this product. We have not tested the Smad7 Antibody (B-8): sc-365846 on mouse kidney tissues. The molecular weight of murine mothers against decapentaplegic homolog 7 protein is approximately 46 kDa. We would expect to obtain bands at this size between 45 - 50 kDa on a Western Blot. We do not have any information about the band sizes you described. Rated 1 out of 5 by from Not appropriate for tissue extractsDidn't work with extracts from skeletal muscles overexpressing human Smad7. I used the recommended titer of 1:100, which is utterly ridiculous, and still detected nothing. Antibodies from other vendors worked (see picture) and I suspect the product has been diluted to the point that it only works with cell culture extracts. This is growing problem in the field and suppliers need to stop doing this; it's better to charge more for a product that works than sell a highly diluted one that doesn't. Seriously, 1:100?!?! SC honored their guarantee though and provided a different antibody for a different protein in "exchange".

Rated 5 out of 5 by from my review for Smad7 (B-8) antibodyI tested a free antibody for Smad7 (B-8) (sc-365846) using mouse fibrotic kidney samples for Western blot. We loaded 80 ug proteins in each slot in the SDS gel. The antibody was diluted in 1:500 in 3% BSA/PBST and incubated overnight with the blot. We can observe a very clear band around 46 kd. We have repeated this experiment several times and used the fresh diluted antibody on the fresh blot.
