caspase-9 p10 Antibody (F-7) is a mouse monoclonal IgG2a kappa light chain antibody that detects the precursor and p10 subunit of caspase-9 of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-caspase-9 p10 antibody (F-7) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Caspase-9 is a crucial component of the apoptotic pathway, functioning as an initiator caspase that activates downstream effector caspases, such as caspase-3 and caspase-7, in response to pro-apoptotic signals. This activation is vital for the execution of programmed cell death, which is essential for maintaining cellular homeostasis and eliminating damaged or unwanted cells. Proper regulation of caspase-9 activity is critical, as dysregulation can lead to various diseases, including cancer, where evasion of apoptosis allows for uncontrolled cell proliferation. Caspase-9 interacts with several proteins, including Apaf-1, which forms the apoptosome complex that facilitates caspase-9 activation in response to cytochrome c release from mitochondria. Understanding these interactions and regulatory mechanisms involving caspase-9 is important for developing therapeutic strategies aimed at modulating apoptosis in disease contexts.

Hello my name is Aidan. I am doing a research project at UT Southwester research center. I am looking into using a western blot and caspase-9 to detect the amount of P10 in the cell but do not know what lysis buffer to use. do you have any recommendation?

Thank you for your question. We do not yet have a specific protocol recommended for detection of the p10 subunit of caspase-9 with this antibody. We use our standard Western blotting protocol for testing this antibody and observe strong and specific signal for the precursor protein. We use our RIPA Lysis buffer. We would recommend checking some of the product citations this antibody for Western blotting, to see if there is any specific recommendation for inducing and detecting the p10 subunit by Western blot.

Can you recommend a positive control to use with caspase-9 p10 (F-7): sc-17784 monoclonal antibody for Western blot?

We recommend Jurkat (sc-2204), K-562 (sc-2203), HCT-116 (sc-364175), Saos-2 (sc-2235), COLO 205 (sc-364177) or HEK 293 (sc-45136) whole cell lysates to use as positive controls as we have great Western blot data showing specific detection of caspase-9 p10 in these cell lines.

I am using the LiCor/Odyssey system. Which secondary antibody do you recommend using with caspase-9 p10 (F-7): sc-17784?

We recommend using one of our our exclusive Mouse IgG Binding Proteins (m-IgGκ) conjugated to CFL 680 or CFL 790. They are suitable for use with Near-Infrared (NIR) detection systems, such as LI-COR/Odyssey and other comparable systems.
