Caspase-9 Antibody (96.1.23) is a mouse monoclonal IgG1 kappa light chain antibody that detects caspase-9 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-caspase-9 antibody (96.1.23) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Caspase-9 plays a crucial role in the apoptotic pathway, acting as an initiator caspase that activates downstream effector caspases, such as caspase-3 and caspase-7, thereby facilitating programmed cell death. This process maintains cellular homeostasis and eliminates damaged or unwanted cells, which is particularly important in cancer and neurodegenerative diseases. Caspase-9 activation is tightly regulated and occurs in response to apoptotic signals, highlighting its significance in cellular response to stress and damage. Understanding caspase-9 function and interactions with other proteins in the apoptotic cascade can provide insights into therapeutic strategies aimed at modulating apoptosis in various diseases.

I cannot detect Pro-caspase 9 and Caspase 9 bands in HT29 cells. I would like to ask what I should use to dilute the antibody (3% BSA or 3% skim milk), what blocking buffer to use (3% BSA or 3% skim milk), the recommended dilution ratio for this cell type

Is this cas9 antibody(caspase-9 抗体 (96.1.23): sc-56076) suitable for immunofluorescence in frozen section of mouse? The protein what I want to detect is exogenous by caspase-9 CRISPR technique. Is this antibody suitable?

Thank you for your question. Yes, caspase-9 抗体 (96.1.23): sc-56076 is suitable for immunofluorescence in frozen section of mouse. This antibody is raised against the prodomain of caspase-9 of human origin, we did not test it on exogenous cas9 by caspase-9 CRISPR technique.

Can this antibody be used to stain mouse tissue in IHC? Are there any mouse on mouse background issues?

Thank you for your question. Yes, this antibody is appropriate for use with mouse tissue. In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This antibody is currently available conjugated to HRP, PE, FITC, AF488 and AF647.
