ZSCAN22 employ various cellular mechanisms to modulate the activity of this protein. Forskolin, for instance, directly stimulates adenylate cyclase, which catalyzes the conversion of ATP to cyclic AMP (cAMP). This increase in cAMP levels leads to the activation of protein kinase A (PKA), which can phosphorylate ZSCAN22, thereby promoting its activation. Similar to forskolin, compounds such as 8-Br-cAMP and Dibutyryl-cAMP are cell-permeable cAMP analogs that also activate PKA, with the same subsequent phosphorylation and activation of ZSCAN22. On the other hand, IBMX works by inhibiting phosphodiesterases, thereby preventing the degradation of cAMP and prolonging the activation of PKA, which again leads to the phosphorylation of ZSCAN22.
Additional compounds activate ZSCAN22 through different signaling pathways. PMA and its derivative Phorbol 12-myristate 13-acetate, both function as activators of protein kinase C (PKC). The activation of PKC leads to the phosphorylation of proteins, and this could include the phosphorylation and resultant activation of ZSCAN22. In a similar vein, Calyculin A serves to maintain proteins in their phosphorylated state by inhibiting serine/threonine phosphatases, thus potentially keeping ZSCAN22 active. Agents such as Ionomycin increase intracellular calcium levels and can activate signaling cascades that result in the activation of proteins like ZSCAN22. The Epidermal Growth Factor (EGF) activates its receptor tyrosine kinase, initiating a cascade that may lead to the activation of the MAPK/ERK pathway, which in turn can phosphorylate ZSCAN22. Similarly, Zinc Pyrithione activates MAPK pathways, leading to the phosphorylation and activation of ZSCAN22. Lastly, Insulin stimulates its receptor to begin a signaling cascade that activates the PI3K/Akt pathway, with Akt capable of phosphorylating several substrates that could be involved in ZSCAN22 activation, and Anisomycin acts as a JNK activator, which can result in the phosphorylation of transcription factors and other proteins that may play a role in the activation of ZSCAN22.
