The involvement of calcium in the activation of XAGE1E is evident with the use of ionomycin, which raises intracellular calcium levels and can activate calcium-dependent kinases that may phosphorylate XAGE1E. Epidermal Growth Factor (EGF) binds to its receptor, leading to the activation of the MAPK/ERK pathway, which can culminate in the phosphorylation of XAGE1E if it is a part of this signaling cascade. Insulin, through its receptor, kicks off the PI3K/Akt pathway, and Akt can phosphorylate various substrates, potentially including XAGE1E. Anisomycin activates the JNK pathway, which might also involve the phosphorylation of XAGE1E. Lastly, the inhibitors of protein phosphatases, Calyculin A and Okadaic Acid, can lead to sustained phosphorylation states by preventing the dephosphorylation of proteins, which may result in prolonged activation of XAGE1E.
