WDR48, also known as WD repeat domain 48, is a versatile protein implicated in a variety of cellular processes. This protein contains WD repeats, which are minimally conserved regions of approximately 40 amino acids typically ending with tryptophan-aspartic acid (W-D) dipeptide. WD repeats are known for forming a beta-propeller structure that facilitates protein-protein interactions, suggesting that WDR48 serves as a platform for the assembly of protein complexes.Functionally, WDR48 has been implicated in the regulation of deubiquitinating enzymes (DUBs), particularly USP1 and USP46, which are known to play a role in DNA damage response and genomic stability. By forming a complex with these DUBs, WDR48 regulates their activity, impacting processes such as the cellular response to DNA damage and the maintenance of ubiquitination levels critical for cell cycle progression and apoptosis.
In addition to its role in the DNA damage response, WDR48 has been associated with autophagy, a cellular degradation pathway that recycles cytoplasmic components and maintains cellular homeostasis. The interaction of WDR48 with the autophagy machinery suggests a role in the regulation of this essential cellular process, although the precise mechanisms remain to be fully elucidated.WDR48's interactions are not limited to DUBs and autophagy-related proteins. It is also involved in the regulation of telomerase, a ribonucleoprotein complex responsible for the maintenance of telomeres, the protective caps at the ends of chromosomes.
