USP36 Activators are a class of compounds that enhance the functional activity of USP36 through direct or indirect interactions. For example, Forskolin, IBMX, BAY 60-6583, Anandamide, Dibutyryl cAMP, and Prostaglandin E2 all work through the cAMP-PKA pathway. They increase the intracellular cAMP levels, leading to the activation of PKA. PKA then phosphorylates proteins in USP36-related pathways, enhancing its functional activity. Other activators, like A23187 and Thapsigargin, act by increasing intracellular calcium levels, which can activate calcium-dependent kinases that phosphorylate proteins in USP36-related pathways.
Phorbol 12-myristate 13-acetate (PMA) and Chelerythrine chloride operate through the Protein Kinase C (PKC) pathway, altering the phosphorylation state of proteins related to USP36(continued from above) and thereby influencing its activity. Despite having opposing effects on PKC, both compounds can affect USP36 activity through their influence on the phosphorylation state of proteins within the same pathway. Genistein prevents the dephosphorylation of proteins within the pathways that USP36 operates within, thereby enhancing its activity. Lastly, L-NAME increases PKA activation and enhances phosphorylation of proteins in USP36-related pathways by reducing nitric oxide levels, a molecule known to inhibit PKA.
