USP36 Inhibitors

USP36 inhibitors encompass a series of chemicals that predominantly target the deubiquitinating enzymes (DUBs) within the ubiquitin-proteasome system (UPS). Given that USP36 is a constituent of the USP family of DUBs, inhibitors with a broad spectrum, such as PR-619, WP1130, and NSC 632839, can directly inhibit its enzymatic activity, and consequently modulate ubiquitin-controlled processes within cells. These broad-acting compounds exhibit an ability to impede multiple DUBs, highlighting their capacity to modulate USP36 function.

Furthermore, compounds like ML323, P22077, HBX 41,108, and P5091, although specifically targeting other USPs like USP1 or USP7, might bear an influence on USP36 due to their overarching DUB inhibitory actions. Additionally, naturally occurring molecules such as 15-Deoxy-Δ-12,14-prostaglandin J2 and Betulinic acid function as USP inhibitors, suggesting their interaction with USP36. Meanwhile, agents like Mitoxantrone and NMS-873, though not uniquely identified as DUB inhibitors, mediate their effects through proteasomal deubiquitinating activities, introducing another indirect regulation mechanism for USP36.
