L-NG-Nitroarginine Methyl Ester (L-NAME) requires hydrolysis of the methyl ester by cellular esterases to become a fully functional inhibitor (L-NNA). L-NAME exhibits some selectivity for inhibition of NOS. It exhibits Ki values of 15 nM, 39 nM, and 4.4 muM for NOS1 (nNOS) (bovine), NOS3 (eNOS) (human), and NOS2 (iNOS) (mouse), respectively. The reported Ki value for the inhibition of NOS2 ranges from 4-65 muM. L-NAME inhibits cGMP formation in endothelial cells with an IC50 of 3.1 muM (in the presence of 30 muM arginine) and reverses the vasodilation effects of acetylcholine in rat aorta rings with an EC50 of 0.54 muM.

What is the stability in PBS or water?

Thank you for your question. sc-200333A (L-NG-Nitroarginine Methyl Ester (L-NAME), CAS 51298-62-5) is supplied as a crystalline solid. A stock solution may be made by dissolving L-NAME in an organic solvent purged with an inert gas. L-NAME is soluble in organic solvents such as methanol, DMSO, and dimethyl formamide (DMF). The solubility of L-NAME in these solvents is approximately 5 mg/ml. Further dilutions of the stock solution into aqueous buffers or isotonic saline should be made prior to performing biological experiments. Ensure that the residual amount of organic solvent is insignificant, since organic solvents may have physiological effects at low concentrations. Organic solvent-free aqueous solutions of L-NAME can be prepared by directly dissolving the crystalline compound in aqueous buffers. The solubility of L-NAME in PBS (pH 7.2) is approximately 30 mg/ml. We do not recommend storing the aqueous solution for more than one day.
