Ionomycin functions as a calcium ionophore and raises intracellular calcium levels, which can activate calcium-dependent kinases. These kinases have the capacity to phosphorylate Trav12d-1, thereby activating it. Phorbol 12-myristate 13-acetate (PMA) is another activator that works by activating protein kinase C (PKC), which then can phosphorylate Trav12d-1.
Okadaic acid and Calyculin A both inhibit protein phosphatases such as PP1 and PP2A, leading to a net increase in the phosphorylation of cellular proteins, which includes the potential activation of Trav12d-1. Anisomycin activates stress-activated protein kinases, which then can phosphorylate Trav12d-1, contributing to its activation. Thapsigargin leads to an increase in cytosolic calcium by inhibiting SERCA, which can activate calcium-sensitive kinases that may then phosphorylate and activate Trav12d-1. A23187, another calcium ionophore, similarly increases intracellular calcium and can activate calcium-dependent kinases that can activate Trav12d-1. Cantharidin inhibits protein phosphatases, which could lead to the activation of kinases that phosphorylate and activate Trav12d-1. Chelerythrine, while being a potent inhibitor of PKC, can lead to the compensatory activation of other pathways that can also result in the phosphorylation and activation of Trav12d-1. Lastly, SP600125, as an inhibitor of JNK, may lead to the activation of alternative kinases that can phosphorylate and activate Trav12d-1, ensuring the protein's activity within the cell is regulated through multiple pathways and mechanisms.
