TMPRSS11E2 Activators

Chemical activators of TMPRSS11E2 include a range of compounds that ensure the proteolytic activity of the protein is maintained and protected from inhibition or degradation by other proteases. Benzamidine serves to activate TMPRSS11E2 by inhibiting competitive serine proteases, thus preventing them from degrading substrates that TMPRSS11E2 would otherwise process. Similarly, Phenylmethylsulfonyl fluoride provides an irreversible block to serine proteases that could otherwise cleave and inactivate TMPRSS11E2, ensuring that TMPRSS11E2 remains active. Nafamostat and Camostat also contribute to the activation of TMPRSS11E2 by inhibiting serine proteases in the vicinity, which might otherwise bind to and degrade TMPRSS11E2 or its substrates. Gabexate acts in a protective role for TMPRSS11E2 by targeting serine proteases that can cleave and deactivate TMPRSS11E2, thus indirectly enhancing TMPRSS11E2 activity.

Aprotinin increases the functional activity of TMPRSS11E2 by inhibiting a spectrum of proteolytic enzymes that could lead to the degradation of TMPRSS11E2, ensuring it remains functionally active. Leupeptin activates TMPRSS11E2 by binding to and inhibiting serine proteases that would reduce TMPRSS11E2 activity through proteolytic degradation. E-64 takes a similar approach by focusing on cysteine proteases, inhibiting them to prevent the cleavage and inactivation of TMPRSS11E2. Pepstatin A supports TMPRSS11E2 activity by selectively inhibiting aspartic proteases, potentially decreasing the competition for TMPRSS11E2's substrates. AEBSF hydrochloride and 4-(2-Aminoethyl) benzenesulfonyl fluoride hydrochloride both preserve the activity of TMPRSS11E2 by irreversibly inhibiting serine proteases that pose a threat to TMPRSS11E2's stability and activity. Lastly, Chymostatin specifically targets chymotrypsin-like serine proteases, thereby activating TMPRSS11E2 by ensuring that the competing proteases are inhibited, which could otherwise lead to TMPRSS11E2 inactivation.
