TMEM167 Activators

TMEM167 activators include a range of chemical compounds that exert their effects on the protein through different biochemical mechanisms. Some activators work by modulating cyclic AMP (cAMP) levels, a pivotal second messenger in numerous signaling pathways within the cell. When cAMP levels rise, the activation of protein kinase A (PKA) often follows, leading to the phosphorylation of downstream proteins, which can include TMEM167. This phosphorylation can alter the protein's activity, potentially enhancing its function. Other activators influence intracellular calcium concentrations, either through direct ionophoric actions or by modulating calcium channels or receptors. The resulting increase in intracellular calcium can activate calcium-dependent protein kinases, which are capable of modulating numerous proteins, including TMEM167. The alteration in calcium homeostasis and subsequent activation of these kinases could lead to an increase in the functional activity of TMEM167, assuming it is regulated by calcium-dependent mechanisms.
