T2R16 Activators

T2R16 Activators comprise a diverse array of chemical compounds that initiate a series of intracellular events resulting in the activation of the T2R16 receptor, which is integral to the detection of bitter compounds. Denatonium Benzoate, recognized as one of the most bitter substances, directly stimulates T2R16, leading to an increase in intracellular calcium critical for taste signaling. Chloroquine and Quinine, both with antimalarial properties and bitter taste profiles, activate T2R16 in a similar fashion, causing gustducin activation and the release of calcium from intracellular stores. Artificial sweetener Saccharin, at high concentrations, and Propylthiouracil (PROP), both activate T2R16, triggering G-protein-mediated signaling cascades that result in TRPM5 channel opening and cell depolarization. Caffeine, a common bitter compound, and Nicotine, an alkaloid, also target T2R16, enhancing the receptor's calcium signaling pathway essential for taste perception.
