Superoxide dismutase 1 (SOD1) inhibitors represent a class of chemical compounds that interact with the SOD1 enzyme to reduce its activity. The SOD1 enzyme plays a key role in the body's defense against oxidative stress, by catalyzing the conversion of superoxide radicals into hydrogen peroxide and molecular oxygen. However, excessive activity or misfolding of this enzyme can lead to a variety of pathological conditions, hence the interest in chemical compounds that can modulate its activity. SOD1 inhibitors can be classified into two broad categories based on their mode of action: direct inhibitors and indirect inhibitors. Direct inhibitors interact with the SOD1 enzyme itself, often by binding to the active site or otherwise altering the enzyme's structure to decrease its functionality.
On the other hand, indirect inhibitors reduce the activity of SOD1 by altering the overall cellular environment or by modulating other pathways that influence the activity of SOD1. For example, antioxidants such as N-acetylcysteine can indirectly inhibit SOD1 by maintaining the redox balance and preventing oxidative damage, thereby reducing the need for SOD1 activity. Similarly, compounds that scavenge free radicals, modulate the Nrf2 pathway, or alter the cellular redox status can also serve as indirect SOD1 inhibitors.
