Further, the elevation of intracellular calcium levels by compounds like Ionomycin and Thapsigargin serves as a signal for the activation of calcium-dependent kinases. These kinases are capable of phosphorylating SMAGP, thereby activating it. Forskolin contributes to the active state of SMAGP by increasing cyclic AMP (cAMP) levels within the cell, which then activates protein kinase A (PKA). PKA is another kinase that can directly phosphorylate SMAGP. Dibutyryl-cAMP mimics the action of cAMP and similarly leads to SMAGP activation through PKA. Anisomycin, by activating stress-activated protein kinases, also leads to the phosphorylation and subsequent activation of SMAGP. Lastly, 1,2-Dioctanoyl-sn-glycerol (DiC8) acts as a DAG mimic, activating PKC, which then phosphorylates SMAGP, and Epidermal Growth Factor (EGF) engages its receptor to initiate a cascade resulting in the activation of kinases that target SMAGP. Each of these chemicals, through their specific action on kinases and intracellular signaling pathways, ensures the activation of SMAGP, thereby playing a role in its functional regulation.
