SerpinA3a Inhibitors

The protein "serine (or cysteine) peptidase inhibitor, clade A, member 3A," commonly referred to as SERPINA3A (following the nomenclature of the serpin family), is a member of the serine protease inhibitor family. This protein plays a crucial role in regulating protease activity, ensuring that the degradation of proteins by serine (and possibly cysteine) proteases is carefully controlled to prevent damage to cells and tissues.SERPINA3A is part of a larger group known as serpins (serine protease inhibitors), which are a broad class of proteins that inhibit serine proteases – enzymes that cleave peptide bonds in proteins, where serine serves as the nucleophilic amino acid at the active site. Serpins are characterized by their ability to undergo a significant conformational change which irreversibly binds to and inactivates their target proteases, a mechanism that is termed a "suicide inhibition."

In its native state, a serpin exists in a metastable form. Upon interaction with a target protease, the serpin undergoes a transition to a more stable conformation, trapping the protease in a covalent complex and thereby inhibiting its enzymatic activity. This mechanism is essential for the regulation of various physiological processes, including blood coagulation, fibrinolysis, inflammation, cell migration, and immune responses.Mutations in SERPINA3A or dysregulation of its activity can have significant physiological implications, often leading to diseases associated with uncontrolled protease activity. For example, aberrant function of serpins can contribute to conditions such as thrombosis, emphysema, and cirrhosis, among others, where proteolytic balance is disrupted.
