RPL24 Inhibitors

Chemical inhibitors of RPL24 can disrupt its function through various mechanisms, each related to the protein's role within the ribosomal complex. Ricin, a well-known ribosome-inactivating protein, inactivates RPL24 by depurinating the large ribosomal subunit, preventing the binding of elongation factors and effectively halting protein synthesis. Similarly, alpha-sarcin cleaves a specific phosphodiester bond in the ribosomal RNA of the large subunit, compromising the structural integrity necessary for RPL24 function within protein synthesis. Cycloheximide, a glutarimide antibiotic, targets the E-site of the 60S ribosomal subunit, which includes RPL24, blocking the translocation step of protein elongation and causing an accumulation of unlinked amino acids.

Anisomycin operates by binding to the ribosome and preventing peptidyl transferase from forming peptide bonds, a process crucial to the role of RPL24. Blasticidin S also targets the peptidyl transferase center but does so by inhibiting the formation of peptide bonds directly. Emetine, an isoquinoline alkaloid, binds to the 40S ribosomal subunit and consequently inhibits the elongation of peptide chains, which indirectly limits the function of the 60S subunit containing RPL24. Harringtonine and homoharringtonine both act by blocking the peptidyl transferase center of the ribosome, where RPL24 is a critical component, thus preventing the initiation of protein synthesis and the elongation of nascent peptides. Neomycin binds to the 30S ribosomal subunit and induces misreading of the mRNA, indirectly affecting the 60S subunit and the associated RPL24 by causing errors in translation. Sparsomycin binds to the 50S subunit and inhibits peptide bond formation, directly impeding the function of RPL24 in the ribosome. Puromycin causes premature chain termination by mimicking the structure of aminoacyl-tRNA, leading to the release of incomplete polypeptide chains from the ribosome, which includes RPL24. Tetracycline binds to the 30S subunit and inhibits the binding of aminoacyl-tRNA, which indirectly hampers the function of RPL24 by preventing proper translation elongation on the 60S subunit. Each of these chemicals interrupts the ribosome's ability to synthesize proteins by specifically targeting functions that are essential for the active contribution of RPL24, leading to its functional inhibition.
