RNase 7 Activators are a diverse range of compounds that can enhance the functional activity of RNase 7 through various signaling pathways, with most eventually converging on the NF-kB pathway. These include Polyinosinic-polycytidylic Acid (Poly I:C), Lipopolysaccharides (LPS), Pam3CSK4, Flagellin, CpG ODN 1826, and Imiquimod, which stimulate different Toll-like receptors (TLRs) leading to the activation of either the MyD88 or the TRIF pathway. This results in NF-kB activation, enhancing the production of antimicrobial peptides, including RNase 7. Resveratrol operates via the Sirtuin 1 pathway, leading to the deacetylation and activation of NF-kB, thereby enhancing the production of RNase 7. Anisomycin activates the JNK pathway, leading to NF-kB activation and enhanced RNase 7 production.
Zymosan A triggers the Dectin-1-Syk pathway, leading to NF-kB activation and enhanced RNase 7 production. The compounds Curcumin and MG-132 inhibit the IkB kinase and the proteasome, respectively, preventing IkB degradation, which results in increased NF-kB activity and enhanced RNase 7 production. Spermidine, meanwhile, activates the mTORC1 pathway, another pathway leading to NF-kB activation and, consequently, enhanced RNase 7 production. These compounds, therefore, act on various elements of cellular signaling, including the Toll-like receptors (TLRs), the Dectin-1 receptor, the JNK pathway, the mTORC1 pathway, and the IkB kinase-proteasome system. While some of these compounds stimulate receptors to activate the NF-kB pathway, others act by inhibiting the degradation of IkB, an inhibitor of NF-kB, leading to an increase in NF-kB activity, which in turn enhances the production of RNase 7.
