RAM Inhibitors

RAM Inhibitors as a class, based on the assumption surrounding Notch signaling, primarily focus on the modulation of the Notch pathway, either directly or indirectly. Most of the chemicals described, such as DAPT, DBZ, and Compound E, achieve their inhibitory role by targeting the γ-secretase enzyme, a pivotal component for Notch receptor activation. By blocking this enzyme, these inhibitors effectively prevent the cleavage and subsequent activation of the Notch receptor, leading to a cascade of effects on downstream signaling components, including those that contain RAM domains.

In addition to γ-secretase-centric mechanisms, there are other inhibitors like FLI-06 and SAHM1 that take alternative routes. For instance, FLI-06 hinders Notch signaling by influencing endocytosis, thereby impacting the availability and functionality of Notch receptors on the cell surface. SAHM1, on the other hand, has been designed to directly impede the Notch transcription complex, thereby dampening the pathway's downstream effects.
