PP2A-γ Inhibitors

Chemical inhibitors of protein phosphatase 2A gamma (PP2A-γ) encompass a range of compounds that disrupt the normal function of this enzyme by preventing the dephosphorylation of its substrates. Okadaic Acid and Calyculin A are potent inhibitors that increase the phosphorylation levels of proteins by inhibiting the activity of PP2A-γ. Similarly, Cantharidin and Tautomycin target both PP1 and PP2A and function by stabilizing the phosphorylated state of proteins. Endothall also acts by inhibiting PP2A-γ, ensuring that proteins within specific signaling pathways remain phosphorylated.

Additionally, Fostriecin selectively inhibits PP2A over PP1, which results in reduced PP2A-γ activity and, consequently, an increased phosphorylation state of proteins typically regulated by this phosphatase. Microcystin-LR, a cyclic heptapeptide, inhibits the protein phosphatases PP1 and PP2A, including PP2A-γ, culminating in the accumulation of phosphorylated proteins that disrupts normal signaling processes. LB-100 is a small molecule that inhibits PP2A-γ by reducing its ability to dephosphorylate substrates. Cerulenin affects PP2A-γ indirectly by altering lipid metabolism and membrane composition, which can disrupt interactions and signaling pathways associated with PP2A-γ. Lastly, Ionomycin raises intracellular calcium levels that can activate kinases, enhancing the phosphorylation of PP2A-γ substrates and leading to its functional inhibition due to the increased phosphorylation state of these proteins.
