Ionomycin, another chemical activator, facilitates a rise in intracellular calcium, which activates Calcium/Calmodulin-dependent Protein Kinase (CaMK); this kinase can then phosphorylate Olr1519, leading to its activation. Thapsigargin contributes to Olr1519 activation by inhibiting the Sarco/Endoplasmic Reticulum Ca2+-ATPase (SERCA), thus causing an increase in cytosolic calcium that activates kinases capable of phosphorylating Olr1519.
Additionally, A23187 (Calcimycin) and BAY K8644 can lead to the activation of Olr1519 through their respective mechanisms of raising intracellular calcium levels. Zinc sulfate, serving as a cofactor, supports the function of kinase enzymes that phosphorylate Olr1519. Lastly, Sodium fluoride's inhibition of serine/threonine phosphatases ensures the sustained phosphorylation and consequent activation of Olr1519.
