Upon entry into the cell, calcium ions can bind to NICE-3, prompting a conformational change that is essential for its activation. Similarly, BAY K8644 specifically targets L-type calcium channels to boost calcium influx, thereby facilitating NICE-3 activation. Forskolin and Isoproterenol, through their distinct receptor targets, ultimately converge on cAMP production. Forskolin directly stimulates adenylyl cyclase to produce cAMP, whereas Isoproterenol binds to beta-adrenergic receptors to initiate a signaling cascade that also leads to cAMP accumulation. The elevated cAMP levels activate Protein Kinase A (PKA), which can then phosphorylate NICE-3, leading to its activation.
