Nicalin Inhibitors

Nicalin inhibitors often target calcium-dependent pathways or specific kinases to modulate the protein's function. For instance, Cyclosporine A inhibits calcineurin, the dephosphorylation of NFAT. Normally, dephosphorylated NFAT can associate with Nicalin in the endoplasmic reticulum, thereby influencing Nicalin's functional roles in this organelle. Similarly, Thapsigargin disrupts the calcium homeostasis by inhibiting the SERCA pump. Nicalin has been reported to mediate calcium homeostasis; hence Thapsigargin's effects culminate in compromised Nicalin function.

Manipulation of kinase pathways also impacts Nicalin. PP2, a Src kinase inhibitor, disrupts the phosphorylation state of Nicalin, affecting its ability to associate with TMEM147. ML-7, which inhibits myosin light chain kinase (MLCK), affects actomyosin contractility, a cellular event in which Nicalin has been implicated. On another front, Alsterpaullone inhibits CDK2, a key player in cell cycle progression. Nicalin is involved in membrane trafficking processes that occur during interphase, a process regulated by CDK2, and thus Alsterpaullone affects this specific Nicalin function. Therefore, the selection of Nicalin inhibitors needs to be pathway-specific to achieve the desired biochemical outcome.
