Ku-70 Activators

Ku-70 activators are chemicals that can indirectly influence the functional activation of the Ku-70 protein, coded by the XRCC6 gene. This class of chemicals utilizes a diverse range of mechanisms to either directly impact Ku-70 or the cellular pathways where it operates. Genistein and Quercetin, for instance, inhibit specific kinases to impact Ku-70 functionality. Genistein inhibits tyrosine kinases, which can lead to enhanced DNA-binding activity of Ku-70 by reducing its phosphorylation. Curcumin and Epigallocatechin gallate modulate NF-kB pathways and other signaling mechanisms that involve Ku-70, thereby influencing its activation status. Resveratrol and Sodium Butyrate have a unique mechanism involving enzymatic modifications like deacetylation and acetylation, which directly influence Ku-70's affinity towards DNA.

Some activators like Etoposide induce conditions that call for Ku-70's involvement, thus indirectly activating it. Etoposide induces DNA breaks, thereby activating Ku-70 as part of the cellular DNA repair mechanisms. ZnSO4, which provides Zinc ions, plays a critical role in enhancing the DNA-binding activity of Ku-70. In the same vein, Sodium Arsenite induces oxidative stress conditions that can activate Ku-70 as part of the cellular protective response.
