Dibutyryl-cAMP, being a cAMP analog, bypasses the cellular mechanisms for cAMP production and directly activates PKA, thereby streamlining the phosphorylation and activation of KLHL35.
PMA mimics the action of diacylglycerol (DAG) and activates Protein Kinase C (PKC). PKC then potentially phosphorylates KLHL35, leading to its activation. Thapsigargin, by inhibiting the SERCA pump, also increases cytosolic calcium, indirectly fostering an environment that facilitates the activation of KLHL35. Moreover, Okadaic Acid and Calyculin A prevent the dephosphorylation of proteins by inhibiting protein phosphatases PP1 and PP2A. This inhibition may lead to a state of hyperphosphorylation, which can include the activation of KLHL35. Anisomycin, by activating stress-activated protein kinases, can indirectly promote the activation of KLHL35 through phosphorylation. Lastly, Spermidine, through its role in modulating NMDA receptor activity and subsequent calcium influx, may influence the activation of calcium-dependent kinases and phosphatases, contributing to the activation of KLHL35.
