ITPA, or inosine triphosphate pyrophosphatase, is an enzyme encoded by the ITPA gene in humans. ITPA specifically hydrolyzes inosine triphosphate (ITP) and other non-standard nucleotide triphosphates like xanthosine triphosphate (XTP) and deoxyinosine triphosphate (dITP) to their respective monophosphate forms and pyrophosphate.
The enzymatic activity of ITPA is essential for the nucleotide sanitization process, as the presence of non-standard nucleotides such as ITP in the DNA or RNA can lead to errors in replication and transcription, potentially causing genetic disorders or contributing to the carcinogenic process. By removing these potentially harmful nucleotides from the cellular nucleotide pool, ITPA helps to maintain genomic stability and ensures the fidelity of cellular replication and transcription mechanisms. Mutations in the ITPA gene can lead to a deficiency in the enzyme's activity, which is associated with an accumulation of ITP in erythrocytes, a condition known as ITPA deficiency. This condition is generally benign but has been associated with adverse drug reactions to certain purine analogues used in antiviral and anticancer therapies.
