IL-27 Activators

Interleukin-27 (IL-27) is a cytokine belonging to the IL-12 family, which plays a critical role in the regulation of immune system responses. This protein is primarily produced by antigen-presenting cells, including macrophages and dendritic cells. IL-27 is composed of two subunits, EBI3 (Epstein-Barr virus-induced gene 3) and p28, which together form a biologically active heterodimer. The mechanisms governing IL-27 activation and its subsequent effects on immune cells are intricate and highly regulated. Activation of IL-27 involves its interaction with a specific receptor complex composed of IL-27Rα (WSX-1) and gp130. Upon binding to its receptor, IL-27 triggers the JAK/STAT signaling pathway, particularly activating STAT1 and STAT3 transcription factors. These factors then translocate to the nucleus, where they influence the expression of genes involved in immune regulation, including those that promote the differentiation of naïve T cells into Th1 cells and regulatory T cells. This signaling cascade underscores the critical role of IL-27 in bridging innate and adaptive immunity, facilitating rapid and coordinated responses to pathogens. Additionally, IL-27 influences the production of cytokines and chemokines, further modulating the immune landscape.
